epiCS-M / Human Epidermis Equivalent with Melanocytes

The development of new formulations for skin bleaching, tanning or UV-protection is an important challenge for the cosmetic and pharmaceutical industry. Active ingredients used in these formulations have to be tested for efficacy and product safety.

The use of time consuming and expensive animal testing is often a legitimate subject for ethical debates. On the other hand currently applied monolayer cell cultures have several limitations, e.g. requirement of vehicles for hydrophobic substances, lack of a physiological barrier function, and also, the topical application of substances onto the cells is not possible. Organotypic 3-D cell culture systems are the most effective tools to overcome these limitations.

 epiCS-M is specially designed for these needs

epiCS-M is an in vitro reconstructed human epidermis consisting of normal primary epidermal keratinocytes and melanocytes. This pigmented epidermis is produced in cell culture inserts (0.6 cm2) on a polycarbonate membrane at the air-liquid interface. The appropriate culture media does not contain artificial melanogenesis stimulating or inhibiting agents.

This highly differentiated epidermis equivalent shows epithelial stratification and cornification. Melanocytes undergo spontaneous melanogenesis during culture. Immunohistochemical analysis shows physiological numbers of melanocytes located in the stratum basale.

During a culture period of up to three weeks a physiological increase of pigmentation will be observed macroscopically without compromising the epidermal structure. Substances can be applied topically and systemically.  Bleaching experiments with e.g. kojic acid result in a significant decrease of visible pigmentation and melanin content, while tanning experiments with IBMX, alpha-MSH or UV irradiation show strong stimulation of melanogenesis.

epiCS-M is available with melanocytes from three different donor types: Caucasian, Asian-Caucasian or Afro-American. All donor types show the phototype specific pigmentation pattern.

epiCS-M is a useful tool for efficacy testing of substances and formulations within the scope of skin tanning, skin bleaching or UV-protection in vitro.
